摘要:中性粒細(xì)胞是抵御病原體最豐富和最有效的防御者,在不同癌癥類型中發(fā)揮相反的功能。然而,由于它們的半衰期較短,探索中性粒細(xì)胞如何在癌癥中采取特定的命運(yùn)仍然具有挑戰(zhàn)性。在這里,作者從17種癌癥類型(來(lái)自143例患者的225個(gè)樣本)中生成并整合了單細(xì)胞中性粒細(xì)胞轉(zhuǎn)錄組。中性粒細(xì)胞表現(xiàn)出異常的復(fù)雜性,有10種不同的狀態(tài),包括炎癥、血管生成和抗原呈遞。值得注意的是,在大多數(shù)癌癥中,抗原-呈遞程序與良好的生存率相關(guān),并可通過(guò)亮氨酸代謝和隨后的組蛋白H3K27ac修飾引起。這些中性粒細(xì)胞可以進(jìn)一步引起(neo)抗原特異性和抗原非依賴性T細(xì)胞反應(yīng)。中性粒細(xì)胞傳遞或亮氨酸飲食微調(diào)免疫平衡,以增強(qiáng)抗PD-1治療??傊@些數(shù)據(jù)不僅表明了不同癌癥間中性粒細(xì)胞的差異,而且還提示了治療機(jī)會(huì),如抗原呈遞中性粒細(xì)胞傳遞。
