【背景介紹】內(nèi)質(zhì)網(wǎng) (ER) 膜蛋白復合物亞基 10 (EMC10) 與肥胖有關(guān)。作者研究了 EMC10 的兩種同工型,包括分泌型同工型(scEMC10)和 ER 膜結(jié)合同工型(mEMC10),在代謝功能障礙相關(guān)的脂肪肝病 (MASLD) 中的作用。


【研究方法】采用多種脂肪變性小鼠模型和 HepG2 細胞來研究 EMC10 在肝臟 PERK-eIF2α-ATF4 信號傳導和肝脂肪變性調(diào)節(jié)中的作用。探討scEMC10中和抗體對小鼠肝脂肪變性的治療作用。


【研究結(jié)果】scEMC10 促進肝臟 PERK-eIF2α-ATF4信號傳導的激活,而mEMC10 抑制?;蚯贸觿×诵∈蟮母闻K ER 應(yīng)激和脂肪變性,而mEMC10的肝臟過度表達則受到缺乏蛋氨酸和膽堿的飲食或衣霉素的挑戰(zhàn),這突顯了mEMC10通過調(diào)節(jié)肝臟 ER 應(yīng)激在 MASLD 中的直接抑制作用。scEMC10 的過度表達促進了脂肪肝小鼠的肝脂肪變性,而循環(huán) scEMC10 的中和則阻止了其發(fā)生,這表明scEMC10在MASLD發(fā)展中的作用。

For transfection of RFP-GFP-LC3 lentivirus, HL-1 cells were seeded in a six-well plate or 96-well plates and transfected with RFP-GFP-LC3 (Genomeditech Co., Ltd. (Shanghai, China)) following the manufacture's protocol. For transfection of lentivirus, NMCMs or HL-1 cells were seed in 6 cm dishes (for immunoprecipitation), 6-well plates (for immunoblots) or 96-well plates (for immunoflurorences) before transfecting with GST-Rilp or Rab7Y183A lentivirus following protocols from Genomeditech Co., Ltd. (Shanghai, China).
